There are many, many new features and improvements, including latest versions of all core programs, additions to the UberStudent suite of administration tools, improved language support including full language support during installation, and much, much more!
NOTE: upgrading from UberStudent 4.0 or 4.1 will not upgrade your kernel, xorg, or mesa. You should normally avoid upgrading those, unless you have a specific reason related to hardware functionality. Earlier UberStudent 4.* editions will otherwise upgrade to 4.3 just by doing normal updates with the Update Manager. If you want the newer versions of xorg and mesa, which have been backported from Ubuntu 15.04 to 14.04, it is preferable to install 4.3 from the ISO.
